The sustainable composition of diets of high nutritional value is of the utmost importance for intensive aquaculture. Digestion and absorption of nutrients depends on the activity of the digestive enzymes, in particular those located in the brush border membrane of enterocytes, which are responsible for the final stages of breaking down and absorption of nutrients. The enzyme glucokinase (GK) (EC 2.7.1.1) plays an important role in the control of glucose homeostasis. Qualitative and/or quantitative variations in GK enzyme have been postulated by previous studies to explain why dietary carbohydrate utilisation is lower in gilthead seabream (Sparus aurata) and rainbow trout (Oncorhynchus mykiss) than in common carp (Cyprinus carpio). In this study, we report the isolation and characterisation of a full-length cDNA coding for GK in these teleosts. Amino acid sequences derived from these cDNA clones are highly similar to other vertebrate GKs. For developing efficient diets, two sets of experiments examined whether the use and allocation of dietary protein can be traced by labelling with stable isotopes (N-15 and C-13) in two culture fish (Oncorhynchus mykiss and Sparus aurata). In the first experiment, natural abundance and tissue distribution of these isotopes were determined, by measuring the delta C-13 and delta N-15 values by isotopic ratio mass spectrometry, in fingerlings (14-17 g) adapted to diets differing in the percentage of fish meal replacement by plant protein sources. The common dentex (Dentex dentex) is a sparid fish which is considered a suitable candidate for Mediterranean aquaculture. A seven-weeks feeding trial was conducted over the summer period with common dentex, sea bream (Sparus aurata) and sea bass (Dicentrarchus labrax). All three species were fed to visual satiety with two practical diets with varying crude protein and crude fat levels (55 % protein, 9 % lipids; 46 % protein, 17 % lipids). Three practical isoproteic (54% protein) diets were formulated to contain graded levels of crude fat (diet L: 10%, diet M: 20% and diet H: 30%). Each diet was assigned unrestrictedly to three and restrictedly to two replicate groups of fish (IBW 243 g). In unrestricted groups, increasing the dietary lipid level led to a significant decrease in voluntary feed intake without affecting growth rate. High energy extruded diets were formulated to contain the same level of protein supplied either by soy protein concentrate (SPC) or fish meal. Three experiments were performed in order to measure voluntary feed intake and feed waste, faecal losses and soluble losses of nitrogen and phosphorus in rainbow trout (average body weight: 100 g).
